* Ability to make molarity calculations for the preparation of solutions and buffers and for diluting, quantifying and dispensing proteic material

What will you do?
Context
Fermentation, protein purification and analytics team (FPPA lab) operating within the Antigen Design function in Research department is mainly involved in the production, purification, analytical and functional characterization of novel proteic vaccine candidates with the perspective of process transfer at manufacturing scale.
The intern will initially be supported by experienced scientists of the laboratory to learn the fermentation and chromatographic techniques necessary for the production and the purification of the antigen of interest.
Project intern description
The project has the objective to exploit the Production of Bacterial Cell Membrane proteins with different approaches with a special focus on cell free-system technology. The candidate will first produce the bacterial Membrane Proteins and after partial purification he/she will evaluate the chemical-physical properties of the produced proteins, manly focusing on their folding, thermal stability and capability to bind specific and functional monoclonal antibodies. Bacterial Membrane Proteins will be selected from Neisseria Gonorreae, Chlamydia Trachomatis and E. coli genomes.
Competencies the intern will develop thanks to the internship experience
Learn and understand the available techniques for Membrane Proteins analytical characterization. In particular, the student will learn how to express in-vitro proteins, chromatographic techniques, SE-HPLC, SDS-page, western blot, quantification methods, thermal stability analysis, SPR and BLI technics. All these techniques will allow him/her to produce and characterize bacterial Membrane Proteins that can be used to solve its structure and consequently better understand the role of the candidate during bacterial infection.
